The ride from Ungarie to Melbourne, which raised over $140,000 to help find a cure for motor neurone disease in its inaugural running in 2018, will include sports night functions at Ariah Park, Lockhart, North Albury, Benalla, Nagambie and Lancefield.
All money from the 750km trek will go toward the Fight MND campaign spearheaded by Essendon champion and MND sufferer Neale Daniher.
Star Melbourne ruckman Max Gawn will be involved at the North Albury and Benalla events, St Kilda midfielder Jack Steele and dual Brownlow medalist Peter Moore at Nagambie, Paralympic gold medalist Michael Milton at Lockhart and Hawthorn duo Isaac Smith and Luke Breust at Ariah Park.
The event is the brainchild of Lockhart cyclist John Fox, and will take a slightly different route this year.
Cyclists can participate in the entire journey or individual legs, with cyclists departing from Daniher's home town Ungarie on March 8 and arriving at the Bombers' spiritual home Windy Hill on March 14.
"A lot of these towns have been pretty heavily affected (by bushfires), the ones we're coming back to are excited to have us back and we're happy to support them," Fox said.
"We always stay at footy clubs along the way and try and support the backers of the footy clubs while we're there.
"The first one we did, a group of us were thinking we should do a big bike ride one day and it was around the time Neale was fairly prominent in the media.
"It went off so well that out of respect for Neale and his family we wanted to start it again from Ungarie, it seems appropriate."
Fox said continued fundraising will also help raise understanding and awareness of the disease.
"I think everyone involved will have some type of connection with someone affected by it," he said.
"Footy clubs are the heart and soul of small communities, and it resonates with a lot of us.
"The four Daniher brothers are going to be there (in Melbourne) to join us for a finishing function, it will be tremendous to be in the same room as them."
Each rider is required to fundraise, and will receive a link to set up a fundraising page online once they are registered.
