Five-eighth Josh Siegwalt and hooker Sam Macklan both accepted one-game bans after being charged following the requested match review following the 34-16 loss to Kangaroos to start the season on Saturday.
Both took the early guilty plea over careless high tackle charges.
It's left co-coach James Hay to reshuffle his plans with both new recruitments coming into key role for Brothers.
"Every team needs to have sure they have some kind of depth at their club as you know it is going to be tested at some stage during the season but round two is earlier than I would have liked," Hay said.
"Club stalwart Zac Wilson will come into the side somewhere to plug that gap and then we'll have to shuffle a few things around.
"We haven't exactly decided what it will look like, but we should be able to make it work."

Brothers are also set to be without Seb Cottam as the Gundagai premiership winner continues to deal with an ankle complaint picked up in the pre-season.
Darryl Charles also isn't expected to play again.
Kangaroos got off to a fast start against Brothers last week, bouncing out to a 28-0 lead at half-time.
Brothers were able to win the second half and it's given Hay some confidence.
"We probably just lost the jump in the first half," he said.
"Having Cade (Price) sin binned and them scoring three tries in those 10 minutes hurt us in the long run.
"We only lost by three tries so I think a lot of the boys walked off very confident we could have won that game.
"We just need to be able to put on an 80-minute, first grade-standard performance."
However Brothers are set for a bigger challenge up against the team who knocked them out of the finals last season.
Young kicked things off with a 54-10 win over Albury last week.
The Cornish brothers were damaging in their first game for the club and Hay is intrigued how to handle their new approach.
"Young look like they have a very exciting and attacking team so defence has to be on point and we have to be ready to show up from the first minute," he said.
"They have a fair few names and faces in the team so we are not going over there with too much knowledge on how they are going to play so it should be fun."
