The inaugural Knights Rescue Rally will be held over May 5-8 ahead of Newcastle's clash with the Canberra Raiders in Wagga.
The event will raise funds for the helicopter service and help the Knights attract fans to the Riverina match.
Participants will travel from Newcastle through regional towns like Mudgee, Bathurst, Sofala, Carcoar, Parkes and West Wyalong.

They will enjoy activities along the way, including visits to wineries, cafes, pubs and a night out in the Elvis capital of Australia, Parkes.
"If you support the Knights and want to help your local rescue helicopter, then this is the event for you," Westpac Rescue Helicopter Service CEO Richard Jones said.
"The fact that participants will venture through many rural communities that could really use the support is an added bonus."
Knights CEO Phil Gardiner said the club was thrilled to be supporting the rally, noting the long-standing community partnership between the two organisations.
"We encourage Knights supporters and those looking for a fantastic trip through regional NSW to get a team of work mates, family or friends together and enter a car into the Knights Rescue Rally and get behind two great local organisations," he said.
Cars of all shapes and sizes are welcome as long the vehicle is registered in NSW.
