The 17-year-old has been on the podium before at state championship level, but finally stepped on the top dais after timing her move to perfection in the sprint to edge out Illawarra's Lillee Pollock.
Brooker was part of a four-rider breakaway which fought it out, and the win follows her victory at the Hanwood Cup in August.
"It was a really tough race, I just held on and had enough left in me for the finish," Brooker said.
"I just got on the right wheel and put myself in the right position, and I was able to get around them.
"It was a technical course and windy which made it difficult. But I was confident as I'd been racing these girls for a while now."
Tolland teammate Myles Stewart also had a strong day, finishing second behind Thomas Bolton in the under-23 men's race.
