The latest sample was collected in Leeton on Monday and follows a positive detection in a previous sample collected on October 18.
On Tuesday, the Murrumbidgee Local Health District said no further test results were available, but confirmation came through late on Wednesday of the second positive detection.
Though there were 34 new cases across the MLHD announced on Wednesday, there are no confirmed COVID-19 cases in the area.
NSW Health also announced positive samples had been found at the treatment plant at Young.
The presence of these fragments could signal undetected cases in the community or that a person with COVID-19 has travelled through the region.

Everyone in the district should be alert for symptoms of COVID-19, including headache, nausea, sore throat, runny nose, cough, loss of smell or taste, or fever.
If symptoms appear, people must isolate and get tested immediately, and remain isolated until a negative result is received.
