Played in recognition of the diggers who used the game as a means of passing time in the trenches, the heads-or-tails based gamble was sorely missed in 2020 amid the pandemic.
This year, most pubs across the city have welcomed it back with open arms.
William Farrer Hotel licensee David Barnhill said pub-goers could try their luck from midday.
"It's going to be a great day, we're all really looking forward to it and with last year cancelled, we're keen to have it back," he said.
"I reckon there will be a lot of Two-up going on in Wagga."

Mr Barnhill hit the mark, with most pubs hosting the game.
Wagga publican Sean O'Hara said The Farmers, Palm and Pawn and The Sporties would each host Two-up from midday.
"They're Wagga's three best beer gardens, so choose your favourite, come along and have a great time," he said.
The Riverina Hotel and Kooringal Hotel will also be holding games on Sunday afternoon, and while the Victoria Hotel won't, it will have Anzac Day entertainment in the form of the band Mobes from 6pm.
While a well-known tradition, not everyone is across the rules. So for those still unsure, here's how to play Two-up:
A nominated 'spinner' throws two coins (preferably pennies) in the air, above head height, from a wooden paddle known as a 'kip'.
One coin is placed heads up, the other tails up.
Players then gamble on whether both coins will land heads-up or tails-up.
The coins must stay within a designated space - off limits to punters.
A 'ringer' oversees the game and ensures fairness.
Winner takes all.
Under the Gambling (Two-up) Act 1998, the game can be legally played on Anzac Day, Victory in the Pacific Day and Remembrance Day (after noon).
