The home side reached 0-210 at tea on Monday with Weatherald unbeaten on 117 and rookie opener Henry Hunt 85 not out, nearing his maiden first-class ton.
After winning the toss and batting, South Australia reached 88 without loss in the first session before picking up the pace after the break.
Weatherald opened his shoulders later in the middle session to reach his ton off 150 balls, with 18 boundaries.
NSW-born Hunt posted his second half-century of the summer, passing his previous best of 75 late in the session.
Tasmania are without regular spearhead Riley Meredith, called up for the Australia A match against Pakistan.
South Australia have yet to register a win after three rounds, while Tasmania sit third with one victory.
