The crash occurred at about 5.30am on the junction of the Hume Highway and Sturt Highway.
"Three males aged 25, 30 and 39 years old are all in a stable condition," said a Murrumbidgee Local Heath District spokesperson.
The 25-year-old driver was transferred to Sydney's St George Hospital this afternoon in a serious but stable condition.
Acting Inspector Geoff Gray, based at the Wagga Police Station, said the car lost control.
"There was a vehicle heading south on the Hume Highway, it came to the intersection of the Sturt Highway and for some reason, it managed to lose control and rolled onto its roof," Acting Inspector Gray said.
"There was some road diversions in place for one to two hours, a 25-year-old male driver was airlifted to Wagga Base and the two passengers were taken by road to Wagga Base Hospital.
"They're all in a stable condition."

Three people have been transferred following a crash on the Hume Highway near Tarcutta following a car crash.
According to Live Traffic NSW, one southbound lane has opened again at the Sturt Highway and southbound motorists can now travel past a car crash.
"The diversion via the Sturt Highway and Olympic Highway has been lifted," a spokesperson said.
"One southbound lane remains closed on the Hume Highway with emergency services and traffic crews still on site.
"There may be intermittent closures on the Sturt Highway as a helicopter is there and motorists should allow extra travel time and exercise caution."
NSW Ambulance Media did not have details of this incident.
Rural Fire Service public liaison officer Bradley Stewart said it was a single vehicle accident.
"It was located at the junction of the Sturt and Hume highways and the accident resulted in short term closure to the Hume," he said.
"The RFS Brigade and the Tarcutta Rural Fire Brigade were at the scene."
