Four occupants were taken to hospital - including two girls aged 10 and 11.
Emergency services were called to the accident about 40km south of the town, near the Kyeamba rest area, at 11.10am.
Wagga police duty officer, Inspector Peter McLay, said a Subaru was northbound on the highway when a Land Rover entered the road from an adjacent rest area.
"In doing so, the Subaru has collided with the rear driver's side of the Land Rover.
"The Subaru then left the road and came to rest on a guard rail."
Inspector McLay said a fire then started in the engine bay of the vehicle, which was quickly extinguished.
A spokeswoman for Ambulance NSW said an 11-year-old was treated for abrasions to her neck, shoulders and abdomen, while another young girl, 10, suffered chest bruising and pelvic pain.
Both were passengers in the Subaru.
A 54-year-old female suffered minor abrasions and another woman, 40, had injuries to her right eye and abdomen.
All were taken to Wagga Base Hospital in a stable condition, while the other two didn't require transport.
The crash forced the closure of one lane of the highway for close to two hours, causing some delays for motorists.
