The closure of the branch in Narrandera has led to two job losses and concerns for elderly access to financial services.
ANZ spokeswoman Emily Kinnear said two employees have accepted redundancy packages, with another to be redeployed elsewhere.
The closest ANZ branch to Narrandera is in Leeton - 25 minutes drive from the town.
The closure follows a "detailed look" at ANZ operations in Narrandera, which found branch patronage had "fallen dramatically over the years".
The availability of electronic banking is blamed for the reduction in physical branch use.
Narrandera Senior Citizens Club secretary Jan Bock said the closure of the branch would force senior citizens to turn to other banks.
Mrs Bock said senior citizens were often not privy to electronic banking and preferred face-to-face interaction.
"The closure will affect older people," she said.
"There are a lot of people that don't know how to use the internet.
"They say you can drive to Leeton to go to the ANZ there, but that is not always easy."
The other "big four" banks - Commonwealth, Westpac and NAB - have branches in the town, as well as smaller players Bendigo Bank and Country First Credit Union.
The ANZ will maintain a presence in the town in the form of an ATM.
The closure of the ANZ branch in Narrandera is the third bank to close in the Riverina this year.
In July, the Westpac branch at Coolamon closed a month after St George departed Cootamundra.
