Connor McCauley will line up in the Southcity second row while younger brother Mitch will do the same for Brothers as the crosstown rivals prepare to renew acquaintances.
They pair played once together in 2019 before taking on each other earlier this season.
This time 19-year-old Mitch will have a bigger role in the starting line up.
Connor made the switch to Southcity this season and is looking forward to the chance to get another one over his little brother and his former teammates.
"I've been giving it to him a little bit throughout the week so hopefully we end up on the same side and can finally see who is the top dog at home," he said.
"He didn't play too much in round one but is starting this week so it will be a good opportunity to go head-to-head."

Southcity scored a 50-0 win when the teams last met in round three.
It's been a tough season for Brothers but Mitch is confident it will be a good learning experience.
"It's a good experience and obviously we would like to even get one win but every club has to go through this and we just need to overcome it," he said.
"It's good playing with the group of boys and we are definitely bonding as a team now.
"We had a lot of new players and a lot of young players but as we train and play together the better we are.
"Even on the weekend not losing to Young by 74 points, only 40, is a little thing."
Connor played four seasons with Brothers before deciding to make the switch this season.
It's a move he's enjoyed.
"It's a strong competitive comp at the moment and no real easy beats so it is always good week-in, week-out to test yourself as a team," Connor said.
"It's been really good winning most of our games and sitting high up on the ladder so I can't complain."
Mitch played made his first grade debut in 2019 before sitting out last season with a knee injury.
He hasn't missed a game yet but has been one of the bright notes for the side who have almost a completely new line this time around.
"Jacob Crossley and Mitch McCauley I just can't praise enough," co-coach James Hay said.
"They are holding down both edges for us and the improvement both have made already is phenomenal."
Brothers head into the clash without Cade Price (hamstring) with Sam Macklan shifting to fullback while Mathew O'Neill will start at halfback.
Tim Hurst will also miss the clash for Southcity with an ankle complaint.
Simon Parr is set to start in the front row.
