The Wagga rivals played out a draw to start the season and only Brooke Wiggett's goal-kicking separated the two times at Equex Centre in the rematch.
Wiggett was able to convert Madison Dunn's try close to the posts to complete the come-from-behind win.
Down on numbers, coach Chris Suckling was pleased with the 12-10 win.
"We got there in the end and that's the main thing," Suckling said.
"It was a good effort from the girls considering we only had one sub."

Kangaroos took advantage of a couple of defensive lapses to score through Lauren Jolliffe and Molly Antone.
However Suckling was pleased with how the team responded to keep their unbeaten record intact.
"We had to hang on but we defended pretty well," he said.
"Two missed tags led to two tries. It's just those one-on-one missed tags that you have to make."
With plenty of players missing, Suckling was impressed with how Ella Clark stepped up while Kirsty Blay, who scored the first try for Brothers, and Emma Suckling split the players player honour.
Suckling believes there is plenty of improvement left in the side who sit one point clear of Temora on top of the table.
"We just have to keep working hard," he said.
The Dragons sit one point behind them after taking a 32-10 win over Junee on Sunday.
Ruby Watterson crossed for a double in the win.
Meanwhile Albury added to their strong start after overcoming a challenge from Young at Alfred Oval on Sunday.
The premiers only have one win to their credit, but once again pushed a top four rival before slipping to a 12-6 loss.
They will be looking to hit back when they face Brothers at Equex Centre on Sunday.
Meanwhile Southcity and Gundagai will be chasing wins in their battle for fifth place.
The Bulls head south to face Albury while the Tigers host the winless Diesels.
Both teams are locked on 11 points, two behind fourth-placed Kangaroos, after the bye.
