Hopper polled seven votes in the Greater Western Sydney's win over Carlton on Saturday night to overtake Taylor Walker for the lead.
The Carey-Bunton Medal is awarded to the best footballer from NSW in the AFL, based on votes in the AFL Coaches Association Champion Player Award.
The award is run by the NSW Australian Football History Society, in conjunction with the AFL Coaches Association.
Seven votes against the Blues took Hopper to a total of 40 for the season, and put him two in front of Walker, who hails from Broken Hill.
There is a then a gap to Sydney's Callum Mills on 24 votes.
