The withdrawal of Mt Austin High School leaves just five schools to compete for the prestigious cup and forced a change of format.
The pools system of recent years has been abolished, along with semi-finals, with each school to play each other once. The top two teams will then qualify for the grand final on May 19.
The Riverina Anglican College (TRAC) will kick off their Carroll Cup defence at Maher Oval on Wednesday night when they host Kooringal High School at 4.15pm.
After that, traditional powerhouse Kildare Catholic College will meet Wagga High School at 5.45pm.
Wagga High School coach Heath Russell said they will field a young team this year.
"We're going to have a couple of leaders in the team that are probably going to have to take a fair bit of responsibility to direct the young boys around," Russell said.
"We've only got a couple of year 12s and year 11 would be our largest group of players.
"The boys are looking forward to getting back and playing. Even though there was a competition last year, it wasn't the same without a crowd. So being able to represent the school again in front of a crowd, the boys will be looking forward to that."
Blake Walker, Jack Mattingly and Darcy Jolliffe will be among Wagga High's leaders that will take on a Kildare team featuring talented senior footballers like Tom Sase, Sam Emery and Will O'Connor.
Russell expects Kildare to prove a serious challenge.
"It's a bit hard, not knowing a lot about them, but they are a proud school with a long tradition of success," he said.
"We expect nothing less than a really strong team in round one."
