The Cherrypickers led 4-0 for most of the clash before the Wagga side finished too strong at Alfred Oval.
Captain Madison Dunn started the fight back as Brothers scored three tries in 10 minutes to take a 14-4 win.
After a draw to start the season with Kangaroos last week, coach Chris Suckling was pleased with their fight back.
"We did it a little bit tough and were on the back foot for a while with our combinations not working just yet so it was good to come away with the win in the end," Suckling said.
Belle Whitechurch opened the scoring for Young in the 16th minute before Dunn scored in the 38th.
Sophie Crouch then went over seven minutes later to give Brothers the lead before a converted try to Ella Clark sealed the 14-4 win.
With Dunn, Naomi Price and Sarah Carter among the returning faces this season, Suckling is hoping they can guide the new generation of Brothers leaguetag players forward after the club missed a grand final berth for the first time in a decade last season.
"Our young girls stood up and Tippy (Dunn) played a good game as usual and really led by example," Suckling said.
"The young girls are still learning as they step into grade but the older girls are there to calm them down and keep them going."
