It is understood the sedan was in the northbound lanes of one of the city's busiest arterial roads when, around 12.40pm, it ended up on its roof in the centre of the four lanes.
It came to rest near the intersection of Leavenworth Drive, near Mount Austin High School.
A spokesperson for NSW Ambulance confirmed crews were called to the crash site around 12.40pm, where three patients were reported to be conscious and breathing.
A woman sustained a head injury in the crash and has been taken to Wagga Base Hospital, the spokesperson said.
The remaining two patients suffered minor injuries.
Police, paramedics and Fire and Rescue NSW personnel were in attendance, with traffic stopped on both sides of Bourke Street while the scene was cleared.
