Commencing their innings at the start of the session, Tasmania reached 2-83 at tea on day two in pursuit of SA's mammoth 6-490 declared.
Test batsman Matthew Wade is at the crease on 13, with opener Alex Doolan unbeaten on 37.
The Tigers run chase commenced promisingly with Beau Webster and Doolan putting on 64 for the opening wicket before losing two quick wickets.
Webster was lbw to Wes Agar for 32, while George Bailey was caught at the wicket off the bowling of Joe Mennie for 1.
Earlier, SA opener Jake Weatherald narrowly missed a maiden first-class double century, adding just three runs in the morning to fall for 198 after the home side resumed at 1-359.
The main contributor for the Redbacks on day two was wicketkeeper Harry Nielsen, who made the most of a rare opportunity with a quickfire unbeaten 64 off 80 balls.
