Mick Duncan (back), Guy Orton (neck) and Sam Daniel (ribs) have been ruled out of the clash against Wagga Tigers at Narrandera Sportsground.
Griffith coach Will Griggs, who has been ill this week as well, is confident they can overcome the loss of the three players.
"We didn't have Orton on the weekend, Mick really only played five minutes and Sam Daniel didn't play the second half so it doesn't really matter who's there, I think we can shake things around to manage without them," Griggs said.
Theo Valeri returns from injury for Griffith, while the Swans have also promoted Michael Agnew, Jordan Crowe and Sam McGinn.
It will be McGinn's first senior game of the year, while Agnew has been called up for his first senior game since June.
Alex Overs was not named in Griffith's team by mistake but will play.
