Emergency services were called to the major highway shortly after 7.30am on Thursday, responding to reports a car had crashed into a tree and rolled at Mullengandra.
A man, who had been travelling in the front passenger seat, died at the scene.
Three male passengers were found trapped in the vehicle and had to be freed from the wreck.
The driver, a man, was taken to Albury Base Hospital in a critical condition.
Two of the passengers have also been taken to hospital by road ambulance, while a third will be airlifted from the scene.
The ages of all involved are unknown at this stage, police said.
A spokesperson for NSW Ambulance said seven crews, including a rescue helicopter, were tasked the crash around 7.35am.
Murray River Police District officers are still on the scene and an investigation into the incident has been launched.
Motorists are urged to exercise caution and reduce speed in the area.
One southbound lane of the highway has been closed as emergency services work to clear the scene, a spokesperson for the NSW Transport Management Centre advised.
Drivers should expect a slight delay as they pass the crash site.
"One of two southbound lanes is closed, so one lane is still open," they said.
"It is quite a serious crash so probably will take some time to clear completely."
Updates to come.
