Hopper was close to returning for Greater Western Sydney (GWS) after undergoing finger surgery but rolled his ankle at training last weekend.
GWS head of athletic performance David Joyce said it was a frustrating blow for Hopper.
“This is an unfortunate setback for Jacob, who had done an enormous amount of work in recent weeks,” Joyce said.
“He has demonstrated his resilience in the past and we’re confident he will attack the rehab plan for this injury with his normal dedication and professionalism.
“We look forward to seeing him back playing an important role for us in the weeks to come.”
Hopper, 20, is out of contract at season’s end and is in hot demand from a number of AFL clubs.
