Funds raised from the day will benefit the Children's Medical Research Institute (CMRI), which works to advance research and treatments in a range of childhood illnesses.
CMRI Wagga committee president Fiona Hamilton said the day had traditionally been well supported in the Riverina.
"We just hope everybody dons their jeans for the day and gets behind some of the stalls they will see and merchandise boxes they will see throughout the Riverina," she said.
"We'd really love the community to continue supporting it - the Riverina is a huge and significant supporter of the CMRI."
Businesses across the city will have Jeans for Genes day merchandise on sale, such as pens, badges and bracelets.
The day isn't just about fund-raising though, according to Ms Hamilton, but about raising awareness for the broader cause.
"It's a national day of recognition for the work scientists and researchers do in researching childhood diseases and creating cures and ways to deal with these things," she said.
